This agreement follows an agreement which saw NOVA use publicly-available knowledge and data to create a bedrock predictive disease model for lung adenocarcinoma which was then validated using an independent database from Paz-Ares1 study (Lux-Lung7 cohort) and benchmarked using the Bayesian model developed by Nagase et al. in 20202.

The companies will now work together to use the JINKO platform to add layers of biological complexity to the initial disease model and then run in silico trial simulations.

"The initial proof-of-concept success means Janssen’s leap-of-faith into in silico has been justified, which makes us very proud," said François-Henri Boissel, CEO. "We are looking forward to deploying JINKO and developing disease models that will provide real insight for drugmakers, streamlining clinical development and getting life-saving treatments to patients faster."

"Janssen is dedicated to employing the most advanced and innovative technologies to ensure new treatments progress through clinical development as efficiently as possible so patients benefit from approved medicines without unnecessary delay. NOVA’s generated proof-of-concept of its predictive disease models through our preliminary work over the past year and we look forward to continuing our ground-breaking work." said Véronique Schmitt, Director of Market Access, Governmental Affairs & Early Products Development – Janssen France.

NEC has partnered with Ono Pharmaceutical to develop AI technology to detect particles in glass bottles

On December 16, 2021 NEC Corporation (NEC; TSE: 6701) reported the development of an AI technology with Ono Pharmaceutical Co., Ltd. that detects particles in glass bottles (Press release, NEC, DEC 16, 2021, View Source [SID1234597515]). This technology uses AI to learn the difference in movements between bubbles and particles in liquid for injections within glass bottles. The technology is even able to detect micro-particles that have been difficult to identify until now with high accuracy. Both companies aim to commercialize the technology in the 2022 fiscal year.

In the process of inspecting liquids for injections that are contained in glass bottles, such as anti-cancer drugs and vaccines, it is common for bottles suspected of containing particles to be automatically removed by inspection equipment, and for a secondary visual inspection to take place by inspectors. Initially, an image-based inspection is performed on all glass bottles. However, normal bottles are often recognized as defective and eliminated. This is due to bubbles being recognized as particles. In addition, advanced skill and know-how are required for inspectors to perform secondary visual inspections, inspection quality often varies, and securing inspectors at the time of production is a challenge.

NEC has partnered with Ono Pharmaceutical to reduce the number of visual inspections by applying this new AI technology to automated inspections, aiming to realize rapid processes that feature uniform inspection quality and help to support the expansion of production.

Image of technology utilization
Technology Features
1. Detect particles with high accuracy based on inspection procedures by inspectors
Visual inspection by inspectors involves tilting glass bottles and stirring liquid in order to identify the differences in the movement between particles and bubbles. This technology uses a high-speed camera to capture the inside state of the bottle. Then, it is analyzed as slow-motion video in order to accurately understand the movement of particles and bubbles, and to recognize the slight differences. This makes it possible to detect particles with high accuracy.

2. Various types of micro-particles can be detected by focusing on motion
In recent years, AI that is capable of recognizing objects within images by using machine learning technologies, such as deep learning, is attracting attention. However, even if a system learns to see particles in an image, it is not possible for the system to work for non-learned particles. Also, it is impractical to enlarge the image so that small particles and bubbles can be distinguished. This new technology distinguishes them by focusing on the movement in which the specific gravity and shape features of particles appear. This allows the technology to work for various kinds of tiny particles.

Going forward, NEC will continue to collaborate with Ono Pharmaceutical and to proceed with verification activities, aiming to bring inspection equipment using this technology into practical use.

The World Health Organization reported in 2020 that approximately 37.6 million people globally were living with HIV and an estimated 1.5 million individuals were newly diagnosed as HIV-positive. There are limited methods available to prevent transmission of HIV, and no products are approved to simultaneously prevent pregnancy, STIs and HIV.

This collaboration will focus on determining compatibility and stability of OB-002 in Phexxi and is expected to yield results in the third quarter of 2022. Assuming positive results, the companies will seek government and philanthropic funding for subsequent clinical trials of the MPT vaginal gel product candidate.

"For too long, women have had to assemble a grab-bag of products to protect against pregnancy, various STIs, and HIV," said Saundra Pelletier, Chief Executive Officer of Evofem Biosciences. "This partnership with Orion aims to develop a first-of-its-kind product with potential to revolutionize the way women protect their sexual and reproductive health: no unintended pregnancies, no STIs, and no HIV — all in one applicator."

Ian McGowan, Chief Medical Officer of Orion Biotechnology, added, "It is unacceptable that we are still seeing high rates of new HIV infections among women each year and there is clearly a compelling need to expand the HIV prevention portfolio. Orion is very excited to be collaborating with Evofem to address this important issue and develop an MPT product for women across the world."

In trials to date of Orion’s novel CCR5 antagonist, OB-002 has demonstrated best-in-class in vitro potency. OB-002 was 100% effective in blocking transmission of HIV infection in a non-human primate model.1 A 15-subject Phase 1 study of vaginally administered OB-002 gel demonstrated that the OB-002 gel was safe and well tolerated, with no evidence of systemic absorption.2

Phexxi is a hormone-free, on-demand prescription contraceptive vaginal gel that was launched in the U.S. in September 2020. Top-line data is expected from Evofem’s pivotal Phase 3 trial for the prevention of chlamydia and gonorrhea in women in the second half of 2022.

In addition to potentially being useful for selecting patients who may respond to DB107 therapy, the utility of this genetic biomarker may extend to the broader RRV platform. This RRV platform has the capability of carrying a wide variety of genes, in addition to the gene carried by DB107. Furthermore, this finding demonstrates that Denovo’s biomarker discovery platform has broad utility for such disparate drug modalities as small molecules and viral vectors.

Denovo’s biomarker platform has utility for disparate drug modalities such as small molecules and viral vectors.

"Biomarker-guided DB107 treatment brings a much needed treatment option to patients with recurrent high-grade glioma, a major unmet medical need with median survival less than a year," said Wen Luo, PhD, Denovo’s Chief Executive Officer and Chief Scientific Officer. "Denovo now has three biomarker-guided programs in active development using biomarkers discovered using its biomarker platform, which once again shows that it can deliver repeatable success."

About Denovo’s RRV Platform and DB107
DB107 is an innovative approach utilizing a proprietary gene therapy platform, RRV, combined with a prodrug, to selectively infect and kill cancer cells while stimulating a robust and durable anti-cancer immune response against a tumor with minimal toxicity. DB107 has been tested clinically in solid tumors including recurrent high-grade GBM and colorectal cancer, most recently in a randomized 403-patient Phase 3 trial. DB107 received Orphan Drug Designation in GBM from the FDA and EMA, and Fast Track Designation from the FDA.

About HGG and Glioblastoma
Most common type of HGG is Glioblastoma multiforme (GBM), which is also the most common type of adult primary malignant brain cancer, with 18,000 newly-diagnosed patients in the US and 13,000 deaths annually. Standard treatment for patients with newly diagnosed GBM can include surgery followed by radiation and chemotherapy, but treatment options are limited. The five-year survival rate of patients with GBM is less than five percent.
